Library amplification is required if Tru-Seq-compatible indexing primers are used in the library preparation. However, if library amplification is not performed (i.e., when doing a PCR-free workflow with ≥ 100 ng of library input), Tru-Seq-compatible full-length Y adapters must be used for adapter ligation. When Tru-Seq-compatible full-length Y adapters are used in the library prep, library amplification may or may not be required, depending on the amount of library needed for your downstream application.
